What is being measured? Is it 15% of the substance making up the trichomes? Is it effected by the density of trichomes on the plant? In which case a very frosty 12% strain will be more potent than a less dense 15% strain. Stupid question?

15% of the cannabinoid profile as a whole is THC.

In very potent strains, carefully prepared marijuana can be 30 percent delta-9 THC by dry weight (seeds and stems removed from flowering buds). Buds are the popular name given to masses of female flowers that form distinct clusters.
 
percentage is % of the weight of the sample submitted. So if you submit a 100g sample and it comes back as 22%, you have 22g of THC in the sample.
